You can backup your Site Collection by this steps:
1. Open new Terminal window.
2. Go to:
C:Program FilesCommon FilesMicrosoft SharedWeb Server Extensions14BIN
3. Execute the following:
ststsadm -o backup -url http://yourSite.com -filename C:oneFolderanotherFolderyourBackupFile
